No, these proteins are manufactured in brain cells but they can become mis-folded, causing them to build up and stick together because the enzymes that would normally break them down can no longer fit around them properly.

Science probably will never answer that question.

I think it is similar to asking what is north of north. From our current understanding, time began at the big bang and what existed before is undefined. We couldn't measure it if we wanted. If the universe really has existed forever, obviously, that won't dissuade the question either as humans have such an inadequate grasp on infinity.
 
I have an easier time wrapping my head around an infinite timeline for the universe than I do a singularity before which nothing existed.
