Grady Wilson on Sanford and Son. Minor character that developed into a major character. "Good goobily goop!"

And speaking of Bryan Cranston, Tim Sacksky on King of Queens. "It was liquid heaven!"
 
Lots of good ones already named. Dr. Spaceman, Raffi, the lawyer from Almost Sunny and Tammy #1 from P&R are definitely on my list.

I'll add Dr. Nick, the Sea Captain and of course Ralph Wiggum from The Simpsons. I'd put Hank Scorpio on there, but he's a one-time character.
